- Re-epithelialization after cutaneous injury is a complex and multifaceted process that incorporates numerous cellular components interacting in a myriad of pathways. One of the most crucial aspects of this process is the initiation of keratinocyte migration to fill the wound bed. Re-epithelialization involves both the individual and collective movement of epidermal cells under the control of integrated signaling paradigms. It is therefore essential to develop a simple methodology to dissect the basic movement of epidermal cells in vitro. Scratch assays are relatively simple experiments in which a single layer of cells are plated onto a prepared dish with multiple furrows created in the cell bed. The resulting cellular migration to fill the wound bed can then be imaged and processed quantitatively to investigate migration rates and other factors of interest. Here, we provide important adaptations to the classic scratch assay to make it a robust, reproducible, and quantitative tool for the evaluation of epidermal cell migration.

- Cutaneous wound healing is an intricate and multifaceted process. Despite these complexities, the distinct phases of wound healing provide a unique opportunity to evaluate the roles of different targets in these coordinated responses. This protocol details an in vivo wound healing assay to study the intersection of cellular, molecular, and systemic effector pathways. The role of certain proteins in the wound healing process can be efficiently explored in vivo through the generation of tissue-specific deficient mice. This approach, although optimized for use with animal models displaying epithelial deficiencies, can be used for other tissue-specific deficiencies, and utilizes simple and cost-effective methods, allowing investigators to precisely devise their experimental design. The coordination of immunological, epithelial, vascular, and microenvironmental factors in wound healing makes this technique a valuable tool for investigators across fields.

- Klotho is a regulatory anti-aging protein that is significantly expressed in the choroid plexus, plays an important role in regulating the immune response in the CNS, and diminishes with age. It is known that a decline of klotho expression has been linked to significant neuroinflammation, oxidative damage in the neural parenchyma, and decline in neurocognitive functions. As the choroid plexus is a known reservoir for latent Human Immunodeficiency Virus (HIV) infection, the relationship between HIV and klotho expression could be a cause of the neurocognitive dysfunction that accompanies HIV infections. The objective of this project was to observe levels of klotho expression in the choroid plexus in a Simian Immunodeficiency Virus (SIV) model of HIV infection, as well as to determine a pattern of age-related decline in klotho expression. To carry out the study, Choroid plexus samples of Rhesus macaques of various ages and SIV-infection status were studied. The tissues were stained using immunohistochemistry for polyclonal klotho, structural proteins (vimentin), and nuclei (DAPI), and imaged at varying magnifications of 4x, 10x, and 40x. The images obtained were then analyzed using the fluorescent imaging software Fiji. The epithelial cells of the choroid plexus were measured for their fluorescence intensity to measure their relative klotho expression levels. Although klotho expression was varied in the uninfected Rhesus macaques, there was an overall decreasing trend with the advancement of age. In SIV-infected Rhesus macaques, the choroid plexus had significantly less klotho expression than the uninfected control group regardless of age (p=0.01). Analysis of the uninfected data shows that there was a decreasing pattern of klotho expression in Rhesus macaques. The variability of klotho levels in the uninfected individuals can be attributed to genetic variations between individuals. However, SIV infection significantly reduced klotho expression in the choroid plexus in comparison to uninfected individuals. This decrease in klotho expression was regardless of age. The exact mechanism in which SIV diminishes klotho expression is unknown and additional studies are warranted to examine this phenomenon. - Context: Regional conservation efforts should incorporate fine scale landscape genetic and habitat suitability data for management decisions. This information permits conservation measures to be tailored to a specific landscape. Objectives: We investigated the landscape determinants of gene flow and habitat suitability for the state-threatened Blanding’s turtle (Emydoidea blandingii) in northeastern New York (NNY). We applied the results from each to examine their complementary contributions to local connectivity and genetic structuring. Methods: We conducted population and individual-based genetic analyses with microsatellite data to evaluate genetic structuring and landscape genetics in NNY. We coupled these genetic analyses with species distribution modeling (SDM) to estimate the extent of suitable habitat across this important region for species persistence in the state. Results: Gene flow was strongly associated with open water and cultivated land, indicating the role open water channels play in connecting neighboring activity centers, and the propensity of females to select cultivated land to nest. Species distribution models based on Landsat-derived vegetation indices and percentage of scrub-shrub wetlands accurately identified Blanding’s turtle habitat. Connectivity estimates from our NNY focal area using landscape genetic and SDM resistance surfaces showed potential movement constraints between the two genetic clusters. Conclusions: Land cover better explained genetic distance data than geographic distance for Blanding’s turtles in our focal area. Accurate SDMs were developed for our focal area with a small number of occurrences (< 50). Using both gene flow and habitat-informed resistance surfaces revealed localized connectivity constraints associated with each, permitting more comprehensive landscape planning.

- Lipid metabolism affects cell and physiological functions that mediate animal healthspan and lifespan. Lipidomics approaches in model organisms have allowed us to better understand changes in lipid composition related to age and lifespan. Here, using the model C. elegans, we examine the lipidomes of mutants lacking enzymes critical for sphingolipid metabolism; specifically, we examine acid sphingomyelinase (asm-3), which breaks down sphingomyelin to ceramide, and ceramide synthase (hyl-2), which synthesizes ceramide from sphingosine. Worm asm-3 and hyl-2 mutants have been previously found to be long- and short-lived, respectively. We analyzed longitudinal lipid changes in wild type animals compared to mutants at 1-, 5-, and 10-days of age. We detected over 700 different lipids in several lipid classes. Results indicate that wildtype animals exhibit increased triacylglycerols (TAG) at 10-days compared to 1-day, and decreased lysophoshatidylcholines (LPC). We find that 10-day hyl-2 mutants have elevated total polyunsaturated fatty acids (PUFA) and increased LPCs compared to 10-day wildtype animals. These changes mirror another short- lived model, the daf-16/FOXO transcription factor that is downstream of the insulin-like signaling pathway. In addition, we find that hyl-2 mutants have poor oxidative stress response, supporting a model where mutants with elevated PUFAs may accumulate more oxidative damage. On the other hand, 10-day asm-3 mutants have fewer TAGs. Intriguingly, asm-3 mutants have a similar lipid composition as the long-lived, caloric restriction model eat-2/mAChR mutant. Together, these analyses highlight the utility of lipidomic analyses to characterize metabolic changes during aging in C. elegans.

- The White-tailed Ptarmigan (<em>Lagopus leucurus</em>) is one of the few socially monogamous species within the highly polygynous grouse subfamily (Tetraoninae). I found White-tailed Ptarmigan in Glacier National Park, Montana, to be nearly genetically monogamous. Of 58 chicks with putative fathers identified, three were the result of extra-pair copulations (5%). Three of 18 clutches (17%) contained extra-pair offspring. I suggest that White-tailedPtarmigan males are able to guard their females effectively from extra-paircopulations because of high visibility in their habitat and their ability to forage alongside their mate. The three extra-pair offspring were sired by unknown males.

- AIMS: Recent studies have suggested a possible association between microbiota and gestational diabetes (GDM). However, the results are inconsistent. Our objective was to investigate further the relationship between GDM and microbiota and verify the potential microbial marker. METHODS: Two complementary approaches were used for the demonstration. First, we compared the gut microbial composition of 23 GDM patients and 26 non-GDM ethnically Chinese Han pregnant women, by using whole-metagenome shotgun sequencing of their stool samples collected at the third trimester. Second, we used Q-PCR (quantitative polymerase chain reaction) to evaluate the gut microbial composition in the stool samples from another cohort of 150 Chinese pregnant women (113 Control and 37 GDM), to further confirm the potential microbial marker. RESULTS: The gut microbiota of GDM women show lower albeit not statistically significant (p = 0.18) alpha diversity at the species level than non-GDM women. However, the species-level beta-diversity or between-sample diversity measured by Bray-Curtis distance shows significant differences (p < 2.2e-16) between the two groups. The species Bacteroides dorei positively correlated with both OGTT (oral glucose tolerance test) 0-Hour (p = 0.0099) and OGTT 1-Hour (p = 0.0070). There is a similar trend between Bacteroides sp. 3_1_33FAA and both OGTT 0-Hour (p = 0.014) and OGTT 1-Hour (p = 0.0101) response variables. The species Alistipes putredinis negatively correlated with OGTT 1-Hour (p = 0.0172) and OGTT 2-Hour (p = 0.0147). Q-PCR validation further confirmed the association between the glucose tolerance loci of Bacteroides dorei and OGTT response. CONCLUSIONS: Gut microbiome is related to the diabetic status of Chinese women during pregnancy. Specific species such as Bacteroides dorei associate with glucose response and could be potential monitoring and therapeutic microbial markers for GDM.

- The goal of this research was to determine the effects of the growth of invasive plant Amur Honeysuckle (Lonicera maackii) on the rhizosphere bacterial community composition, and diversity in an urban wetland forest ecosystem. Bacterial communities from the rhizosphere of 5 L. maackii plants and control bulk soils that did not have any L. maackii were investigated at Nina Mason Pulliam EcoLab (NMPE) using a culture-independent pipeline. Bacterial communities were characterized by PCR amplification and cloning 16S rRNA gene fragments following total DNA isolation from the soil samples. Microbial communities associated with both L. maackii rhizosphere and control sites showed high bacterial diversity within each site and taxa unique to individual sites were observed. Phylogenetic analyses revealed 80% of 400 16S rDNA clones were classified as α-, β- and γ-Proteobacteria, Acidobacteria, Actinobacteria, Cytophaga-Flexibacter-Bacteroides (CFB) group, and Verrucomicrobia. Members of the Proteobacteria and Acidobacterium represented 66.5% and 14.5% of the clone library, respectively, whereas the remaining bacterial divisions each comprised less than 7% of the clone library. Twenty-five 16S rDNA clones could not be classified into any known bacterial divisions. Statistical analyses showed significant differences in the presence of L. maackii on the proportions of 16S rDNA clones affiliated with Proteobacteria and Acidobacterium, suggesting bacterial community composition and structure does significantly change in the presence of L. maackii. However, sequence-based community analysis and the corresponding lack of intact microbial cultures limit understanding of the potential influences of enriched microbial taxa on plant hosts and their roles in ecosystem functioning.

- ABSTRACT.—Because of the “montane island” effect and relative rates of climate change, alpine species may be particularly affected by changing climate. White-tailed Ptarmigan (Lagopus leucura) are adapted to life in the alpine cold, but may be less well-adapted to hot summer temperatures. In 1996 and 1997, ptarmigan living in Glacier National Park (GNP) were closely associated with remnant snow and free water in the late summer, a resource that is rapidly changing in distribution and extent as climate changes. We examined White-tailed Ptarmigan distribution, habitat, and numbers in 2009-2010 for comparison with 1996-1997 data. Briefly, five areas within GNP were searched for ptarmigan during August. Habitat data including microclimate were collected at flock locations. Microclimate at ptarmigan flock locations differed from other areas nearby. Flocking ptarmigan appeared to be less numerous than described in 1996 and 1997, and mean flock locations on Logan Pass moved 335 m upslope between the 1990s and 2009 and 2010. Ptarmigan in 2009 and 2010 chose habitat that had the same coverage of vegetation and rock as 1996 and 1997, but contained less snow and water, was farther from snow and water, contained lower soil moisture content, was higher in elevation, and steeper in slope. Our study demonstrates that White-tailed Ptarmigan in GNP have changed distribution, altered habitat preferences, and perhaps on a local scale, experienced declining population numbers in late summer.
